Design: "5-10g genomic DNA was sheared into 10-15 Kb fragments using a g-TUBE device. Then library was constructed using the SMRTbell Express Template Preparation Kit 2.0. Briefly, the sheared fragments were through single-strand overhangs removing, DNA damage repair, end-repair, A-tailing, barcoded overhang adapters ligation. The library was quantified using Qubit 3.0 Fluorometer (Invitrogen, Carlsbad, CA), and the size of the library was checked using an Agilent 2100 Bioanalyzer System. Subsequent steps were followed as per the manufactures instruction to prepare SMRTbell library. The library was sequenced using the Sequel II sequencing platform."
